How exactly does Alzheimer's disease affects neuronal function, in order to cause memory loss?

Disease Alzheimer's is characterized by loss of neurons and synapses in the cerebral cortex and some subcortical regions. This results in atrophy of the gross loss affected regions, including degeneration in the temporal lobe and parietal lobe, and parts of the frontal cortex and cingulate gyrus. Both amyloid plaques and neurofibrillary tangles are clearly microscopically visible in the brains of people affected by Alzheimer's. Plaques are dense deposits, mostly insoluble beta-amyloid peptide and cellular material outside and around neurons. Tangles (NFTs) are aggregates of the microtubule associated protein tau that has become hyperphosphorylated and accumulate within the very cells. Although many older people develop some plaques and tangles, as a consequence of aging, the brain of Alzheimer's patients have a greater number of them in specific brain regions like the temporal lobe.
